A paper on PNAS recently reported the finding of a disease-causing mutation in the -cellenriched MAFA transcription factor by the combination of several approaches. On the basis of exome sequencing result, they revealed that the missense p.Ser64Phe MAFA mutation was associated with either insulinomatosis or diabetes mellitus. Two unrelated families are employed to investigate the relationship between the mutation and the pheonotypes. This is the first time to report MAFA variants might cause disease, exploring the study of mechanisms underlying the regulation on human-cell and tumorigenesis of this transcription factor.
